The impression I'm getting is that killing Lilith is the final seal, and that's why Ruby has been encouraging Sam in that direction. I wouldn't be surprised if the final episode does have Lucifer rising, that way they could have Dean being the one to break the first seal, and Sam breaking the last seal. Season 5 in a post-apocalyptic world would be pretty cool to me...
